What is the best way to turn these leads into buyers or sellers?

First of all, congratulations! I should state right away that I'm a licensed real estate agent, and I work for GuideToRealty.com. In short (and in the interest of full and fair disclosure), I sell leads gathered on the Internet.
The two most important things to have in place before you start to work your leads are (1) contact management software (Outlook is good; ACT! is very good), and (2) attitude. You can find the software easily, and learning to use it well only takes a couple of weeks, so I'm going to focus on number 2 -- the attitude.
Alacrity is important to your success. You should always contact your leads as soon as possible. Use an "autoresponder" to reply automatically if you're showing a house or giving a listing presentation. After an autoresponse is sent, contact the lead as quickly as humanly possible. You can triple or quadruple your rate of success by doing this! I cannot overstate the need to contact leads immediately, and the sad fact is, many real estate agents let a lead "die" before contacting that lead.
